A practical reference on GHRH analogue: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.
Reviewed 2026-06-10. Anything still debated is marked as such rather than presented as settled.
Two related forms appear under the CJC-1295 name. The simpler analogue, commonly written as modified GRF(1-29), carries the four substitutions but no additional conjugation. The second form attaches a maleimidopropionic acid linker to a lysine residue, a modification frequently called the drug affinity complex. That linker reacts with the free thiol on serum albumin to form a covalent bond. Because albumin has a long residence time in blood, the conjugated peptide stays in circulation far longer than the unconjugated analogue.
Receptor level activity follows the canonical GHRH pathway. The peptide binds the GHRH receptor, a class B G protein coupled receptor on somatotroph cells of the anterior pituitary. Binding raises intracellular cyclic AMP, which promotes calcium entry and the release of growth hormone into the bloodstream. Because the molecule acts at the same receptor as the endogenous hormone, its effect is superimposed on the natural pulsatile rhythm rather than replacing it. Whether sustained receptor occupation leads to desensitization is not fully settled.
Downstream of growth hormone, the liver and other tissues increase production of insulin-like growth factor 1, a mediator of many growth-promoting effects. Studies have documented elevated levels of both hormones after dosing, and the rise from the long-acting form persists longer than that produced by shorter-acting analogues. What remains unclear is whether sustained elevation of these markers translates into meaningful clinical benefit, and whether prolonged exposure carries risks that short trials could not detect.
CJC-1295 acts at the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in cyclic AMP and calcium entry, which promotes release of stored growth hormone. Because the peptide mimics the body's own releasing hormone, it amplifies existing secretory pulses rather than driving continuous output. The size of the response therefore depends partly on the subject's own hormonal rhythm and feedback state.
The attached maleimide group explains the unusual duration of the DAC version. After injection it reacts with the thiol of cysteine-34 on serum albumin, forming a stable covalent bond. The resulting conjugate is too large for rapid kidney filtration and is shielded from many peptidases. Reported half-lives for this form reach several days, whereas the version without the group is cleared in roughly half an hour. That gap is the main pharmacological difference between the two.
| Property | Value | Notes |
|---|---|---|
| Peptide class | Synthetic GHRH analogue | Built on a 29 residue sequence |
| Primary target | GHRH receptor (GHRHR) | Class B G protein coupled receptor |
| Unconjugated half life | Minutes | Limited by protease degradation |
| Key modification | Four residue substitutions | Positions 2, 8, 15 and 27 |
| Common synonym | Modified GRF(1-29) | Usually denotes the form without DAC |
The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
The peptide backbone includes a D-alanine at position two, which resists cleavage by dipeptidyl peptidase IV, and several other substitutions that reduce degradation. Its molecular weight is roughly 3.4 kDa without the linker and about 3.6 kDa with it. The molecule is water soluble and is normally supplied as a lyophilized powder. Precise sequence and mass values depend on which variant is described, so technical documents usually state the exact form being referenced.
CJC-1295 is a synthetic peptide designed as a long-acting analogue of growth hormone-releasing hormone (GHRH). Its structure derives from the first 29 amino acids of native GHRH, a fragment often called GRF(1-29). Four substitutions were introduced to slow enzymatic breakdown and extend activity relative to the natural sequence. The compound was developed by ConjuChem as part of a broader effort to improve the pharmacokinetic profile of peptide hormones. It is studied in laboratory and clinical research settings rather than appearing as a naturally occurring substance.
=== Structure activity relationship === The structure of Benzoquinones may explain their toxicity. Benzoquinones are electrophilic and can undergo reactions with nucleophilic groups, for instance on amino acids, altering structure and function of the proteins. Due to the Cl groups on the quinone ring in DCBQ, the overall electrophilicity of the HBQ is increased, which causes higher reactivity with nucleophilic groups, such as glutathione, proteins, and nucleic acids. If DCBQ reacts with glutathione they form conjugates, lowering GSH levels in the cell, which might lead to GSH depletion.

Although for the rest of his career Heseltine's role in Thatcher's downfall earned him enmity from Thatcher's supporters in the Conservative party, this opprobrium was not universal. In a reference to the reluctance of the Cabinet to support Thatcher on the second ballot, Edward Leigh said of Heseltine: "At least he stabbed her in the front". Heseltine believed that he would have defeated Thatcher if she had contested the second ballot. It has been suggested that he should have withdrawn after weakening her on the first ballot, and that he would have been restored to the Cabinet whether or not she continued as prime minister; he writes that this was never seriously suggested at the time.
After hypothyroidism was found to cause a lower basal metabolic rate, this was used as a marker to guide adjustments in therapy in the early 20th century (around 1915). However, a low basal metabolic rate was known to be non-specific, also present in malnutrition. The first laboratory test to help assess thyroid status was the serum protein-bound iodine, which came into use around the 1950s. In 1971, the thyroid stimulating hormone (TSH) radioimmunoassay was developed, which was the most specific marker for assessing thyroid status in patients. Many people who were being treated based on basal metabolic rate, minimizing hypothyroid symptoms, or based on serum protein-bound iodine, were found to have excessive thyroid hormone. The following year, in 1972, a T3 radioimmunoassay was developed, and in 1974, a T4 radioimmunoassay was developed.
=== Metric ounce === A metric ounce is an approximation of the imperial ounce, US dry ounce, or US fluid ounce. These three customary units vary. However, the metric ounce is usually taken as 25 or 30 ml (0.88 or 1.06 imp fl oz; 0.85 or 1.01 US fl oz) when volume is being measured, or in grams when mass is being measured. The US Food and Drug Administration (FDA) defines the "food labeling ounce" as 30.0 ml (1.06 imp fl oz; 1.01 US fl oz), slightly larger than the 29.6 ml (1.04 imp fl oz; 1.00 US fl oz) fluid ounce. Several Dutch units of measurement have been replaced with informal metric equivalents, including the ons or ounce. It originally meant 1⁄16 of a pound, or a little over 30 g (1.1 oz) depending on which definition of the pound was used, but was redefined as 100 g (3.5 oz) when the country metricated.

The most common gastrointestinal complications of anorexia nervosa are delayed stomach emptying and constipation, but also include elevated liver function tests, diarrhea, acute pancreatitis, gastroesophageal reflux disease (GERD), difficulty swallowing, and, rarely, superior mesenteric artery syndrome. Acid exposure from GERD and self-induced vomiting can cause dental problems, such as tooth enamel erosion and gum disease. Delayed stomach emptying, or gastroparesis, often develops following food restriction and weight loss; the most common symptom is bloating with gas and abdominal distension, and often occurs after eating. Other symptoms of gastroparesis include early satiety, fullness, nausea, and vomiting. The symptoms may inhibit efforts at eating and recovery, but can be managed by limiting high-fiber foods, using liquid nutritional supplements, or using metoclopramide to increase emptying of food from the stomach. Gastroparesis generally resolves when weight is regained.
About 89 percent of the bank's $172 billion in deposit liabilities exceeded the maximum insured by the FDIC. Two days after the failure, the FDIC received exceptional authority from the Treasury and announced jointly with other agencies that all depositors would have full access to their funds the next morning. Seeking to auction off all or parts of the bank, the FDIC reopened it on Monday March 13 as a newly organized bridge bank, Silicon Valley Bridge Bank, N.A. Although some characterized the government response as a bailout, the plan did not entail rescuing the bank, its management or shareholders, but rather making uninsured depositors whole from the proceeds of selling the bank's assets, without the use of taxpayer money. The collapse of SVB had significant consequences for startup companies in the U.S. and abroad, with many briefly unable to withdraw money from the bank. Other large technology companies, media companies, and wineries were also affected. For a number of founders and their venture capital backers, this was the bank of choice.
Prolonged esophagitis, particularly from gastric reflux, is one factor thought to play a role in the development of Barrett's esophagus. In this condition, there is metaplasia of the lining of the lower esophagus, which changes from stratified squamous epithelia to simple columnar epithelia. Barrett's esophagus is thought to be one of the main contributors to the development of esophageal cancer.
The unique dissociation patterns help discover the Quaternary structure of the complex. The symmetric charge distribution and dissociation dependence are unique to SID and make the spectra produced distinctive from any other dissociation technique. The SID technique is also applicable to ion-mobility mass spectrometry (IM-MS). Three different methods for this technique include analyzing the characterization of topology, intersubunit connectivity, and the degree of unfolding for protein structure. Analysis of protein structure unfolding is the most commonly used application of the SID technique. For Ion-mobility mass spectrometry (IM-MS), SID is used for dissociation of the source activated precursors of three different types of protein complexes: C-reactive protein (CRP), transthyretin (TTR), and concanavalin A (Con A). This method is used to observe the unfolding degree for each of these complexes. For this observation, SID showed the precursor ions' structures that exist before the collision with the surface. IM-MS utilizes the SID as a direct measure of the conformation for each proteins' subunit. Fourier-transform ion cyclotron resonance are able to provide ultrahigh resolution and high mass accuracy to instruments that take mass measurements. These features make FT-ICR mass spectrometers a useful tool for a wide variety of applications such as several dissociation experiments such as collision-induced dissociation (CID, electron transfer dissociation (ETD), and others.

=== Irrigation === Proper cleansing of a wound is critical in preventing infection and promoting healing of any wound. Irrigation is defined as constant flow of a solution over the surface of a wound. The goal of irrigation is not only to remove debris and potential contaminants from a wound, but also to assist in visual inspection of a wound and hydrate the wound. Irrigation is typically achieved with either a bulb or syringe and needle/catheter. The preferred solution for irrigation is normal saline which is readily accessible in the emergency department, although recent studies have shown no difference in emergency department infection rates when comparing normal saline to potable tap water. Irrigation can also be achieved with a diluted 1% povidone iodine solution, but studies have again shown no difference in infection rates when compared to normal saline. Irrigation with antiseptic solutions, such as non-diluted povidone iodine, chlorhexidine, and hydrogen peroxide is not preferred since these solutions are toxic to tissue and inhibit wound healing. The exact volume of irrigation used will vary depending on the appearance of the wound, although some sources have reported 50–100 mL of irrigation per 1 cm of wound length as a guideline.
The establishment is a modern, structured research institute composed of six laboratories dedicated to Virology, Parasitology, Bacteriology, Medical Entomology, Clinical Biology and Pathology, including a Research Center for animals and a data center. Each laboratory is made up of a dedicated director and staff, including students and international collaborators. Each laboratory has the basic equipment and the space necessary for optimal research. It is available to faculty, students, post-docs and staff from the periphery of the INRB. Due to the structure of the INRB, if sharing and access to individual laboratory equipment is required, access is granted at the request and approval of the directors of these laboratories. The INRB common area includes some major equipment. All collaborators and researchers have, upon request, access to several -80 freezers, liquid nitrogen tanks, centrifuges, water baths, tissue homogenizers, vortexes, incubators, agitators, and all laboratories have access to cold chain equipment such as dry shippers and portable freezers. The INRB was founded in 1984 and has been a World Health Organization collaborating centre since 2018. The INRB and the World Health Organization have worked closely together on research into the effectiveness of the ring vaccination strategy during the 2018 Kivu Ebola outbreak. The National Biomedical Research Institute (INRB) has eight dynamic departments, which participate in the institute's various missions, namely monitoring, research, and training.
Lindow Man, also known as Lindow II and (in jest) as Pete Marsh, is the preserved bog body of a man discovered in a peat bog at Lindow Moss near Wilmslow in Cheshire, North West England. The remains were found on 1 August 1984 by commercial peat cutters. Lindow Man is not the only bog body to have been found in the moss; Lindow Woman was discovered the year before, and other body parts have also been recovered. The find was described as "one of the most significant archaeological discoveries of the 1980s" and caused a media sensation. It helped invigorate the study of British bog bodies, which had previously been neglected. Dating the body has proven problematic, but it is thought that he was deposited into Lindow Moss, face down, sometime between 2 BC and 119 AD, in either the Iron Age or Romano-British period. At the time of death, Lindow Man was a healthy male in his mid-20s, and may have been of high social status as his body shows little evidence of having done heavy or rough physical labour during his lifetime. There has been debate over the reason for his death; his death was violent and perhaps ritualistic. The recovered body has been preserved by freeze-drying and is on permanent display at the British Museum, although it occasionally travels to other venues such as the Manchester Museum.
Historically, blotting paper or cloth were used to extract filter coffee. Modern coffee filters of paper are made from about 100 g/m2 crêped paper. The crêping allows the coffee to flow freely between the filter and the filtration funnel. The raw materials (pulp) for the filter paper are coarse long fiber, often from fast growing trees. For example, Melitta uses up to 60% of bambus in their filters since 1998. Both bleached and unbleached qualities are made. Coffee filters are made in different shapes and sizes to fit into different holders. Most notable are the (paper) coffee filter systems introduced by Melitta (1908, 1932, 1936, 1965), Chemex (1941) and Hario (2004). Important parameters are strength, compatibility, efficiency and capacity. Tea bags also work as a kind of paper filter. They are made from abacá fibers, a very thin and long fiber manilla hemp. Often the paper is augmented with a minor portion of synthetic fibers. The bag paper is very porous and thin and has high wet strength.
In 1935, the name estradiol and the term estrogen were formally established by the Sex Hormone Committee of the Health Organization of the League of Nations; this followed the names estrone (which was initially called theelin, progynon, folliculin, and ketohydroxyestrin) and estriol (initially called theelol and trihydroxyestrin) having been established in 1932 at the first meeting of the International Conference on the Standardization of Sex Hormones in London. Following its discovery, a partial synthesis of estradiol from cholesterol was developed by Inhoffen and Hohlweg in 1940, and a total synthesis was developed by Anner and Miescher in 1948.

The names are often used interchangeably in casual writing, but they are not strictly identical. Modified GRF(1-29) refers to the unconjugated analogue carrying only the four substitutions. The version sold as CJC-1295 with DAC includes the albumin binding linker and clears much more slowly.
DAC stands for drug affinity complex, a term describing the maleimidopropionic acid linker. This group forms a covalent bond with the cysteine residue on serum albumin after administration. The resulting adduct is what extends the circulating half life from minutes to days.
No. It is entirely synthetic and does not correspond to any known endogenous peptide. The design starts from human growth hormone releasing hormone and introduces deliberate substitutions. Any biological effect comes from mimicking the natural hormone at its receptor.
It binds the growth hormone-releasing hormone receptor on pituitary somatotroph cells. Receptor activation raises cyclic AMP and promotes release of stored growth hormone granules. Because the peptide persists longer than natural releasing hormone, stimulation is prolonged rather than brief.
