CJC-1295 is a synthetic peptide belonging to the growth hormone-releasing hormone analog family. It comprises twenty-nine amino acid residues derived from the N-terminal region of natural GHRH. The molecule incorporates several non-natural substitutions that increase resistance to enzymatic degradation. These modifications extend its activity compared with the native hormone fragment. Researchers use it to study pituitary growth hormone secretion in laboratory and clinical settings. This compound is distinct from native GHRH in its stability profile.
Two principal forms appear in the literature and in research supply. One carries a drug affinity complex, a maleimide-based group that forms a covalent bond with circulating albumin. This linkage slows clearance and produces a long-lasting elevation of peptide levels. The other form lacks that group and is often labeled MOD GRF(1-29). It has a much shorter circulation time. Both variants retain the same core receptor-binding sequence. Reported half-lives differ substantially between the two.
At the pituitary, the peptide binds the growth hormone-releasing hormone receptor on somatotroph cells. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone. Somatostatin and other hypothalamic signals modulate this response. Negative feedback from insulin-like growth factor 1 also influences output. The same regulatory architecture operates with the native hormone. Whether the synthetic analog alters feedback dynamics over repeated exposure remains an open question. Most published receptor work uses cell models rather than intact human systems.
Lyophilized peptide powder is comparatively stable when kept dry, cold, and protected from light. Once dissolved, the molecule is vulnerable to deamidation, oxidation, and aggregation, with the rate depending on pH, buffer composition, and temperature. Alkaline conditions and repeated freeze-thaw cycles accelerate loss of the intact peptide. The methionine present in the native sequence is a known oxidation site, which is one reason it was replaced in the modified fragment. Suppliers typically recommend cold storage of solutions and use within a short window.
Analytical confirmation usually relies on reversed-phase high-performance liquid chromatography for purity and on liquid chromatography coupled to mass spectrometry for identity. Mass data reveal the expected molecular mass and can flag truncated or oxidized species. Amino acid analysis and peptide mapping provide sequence-level verification. Immunoassays are used in some biological matrices, but antibodies raised against one releasing-hormone analog may cross-react with another. Reported purity figures depend heavily on the method used, so comparisons between suppliers require matching the analytical approach.
The two variants differ dramatically in how long they persist in circulation. The form lacking the albumin-binding group has a plasma half-life measured in tens of minutes, comparable to the natural hormone fragment. The version carrying the drug affinity complex binds albumin and shows a half-life of roughly six to eight days in human studies. That figure comes from small trials that tracked hormone levels over extended periods. The physiological consequences of sustained versus pulsatile stimulation are still debated and the literature does not settle the point.

Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Naming in this area is inconsistent, and readers should treat product labels with care. In much of the literature and in vendor catalogs, the unqualified term refers to the albumin-binding version, while the version lacking the DAC group appears as modified GRF(1-29), mod GRF(1-29), or the same name with a without-DAC qualifier. Because one abbreviation has been applied to both materials, the only reliable way to identify a sample is to check the stated sequence and the presence of the linker.
CJC-1295 is the name used for a synthetic peptide modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. The compound was described by a Canadian drug discovery company in the mid-2000s as a long-acting research tool. Two closely related molecules share the name in practice: one carries a drug affinity complex, or DAC, group, and one does not. The distinction matters because the two behave differently in circulation.
== Chemistry == Tertiapin peptide is composed of 21 amino acids with the sequence: Ala-Leu-Cys-Asn-Cys-Asn-Arg-Ile-Ile-Ile-Pro-His-Met-Cys-Trp-Lys-Lys-Cys-Gly-Lys-Lys. The methionine residue is sensitive to oxidation, reducing the ability to block the ionic channels. Methionine can be substituted by glutamine in order to prevent the oxidation. The new synthesized peptide is named Tertiapin-Q and does not show any functional change as compared to the original peptide, which makes it a more suitable research tool.
== Adverse effects == Reported adverse events include both local and systemic reactions. Local adverse reactions are characterized by redness, tenderness, and soreness of the skin at the injection site. The most common local reaction is injection site pain. It has been reported to occur in 16% of patients receiving intramuscular injections, and 3% of patients receiving intravenous injections. Less frequently reported side effects include inflammation of veins (1.2%), sometimes associated with a blood clot (3%). The most commonly reported systemic reactions are diarrhea (3%) and rash (2%). Less frequent systemic reactions to ampicillin/sulbactam include chest pain, fatigue, seizure, headache, painful urination, urinary retention, intestinal gas, nausea, vomiting, itching, hairy tongue, tightness in throat, reddening of the skin, nose bleeding, and facial swelling. These are reported to occur in less than 1% of patients.
Three prime untranslated regions (3′UTRs) of mRNAs often contain regulatory sequences that post-transcriptionally cause RNAi. Such 3′-UTRs often contain both binding sites for miRNAs as well as for regulatory proteins. By binding to specific sites within the 3′-UTR, miRNAs can decrease gene expression of various mRNAs by either inhibiting translation or directly causing degradation of the transcript. The 3′-UTR also may have silencer regions that bind repressor proteins that inhibit the expression of a mRNA. The 3′-UTR often contains microRNA response elements (MREs). MREs are sequences to which miRNAs bind, primarily through evolutionarily conserved seed sequences six to eight nucleobases in length. These are prevalent motifs within 3′-UTRs. Among all regulatory motifs within the 3′-UTRs (e.g. including silencer regions), MREs make up about half of the motifs. As of 2023, the miRBase web site, an archive of miRNA sequences and annotations, listed 28,645 entries in 271 biologic species. Of these, 1,917 miRNAs were in annotated human miRNA loci. miRNAs were predicted to have an average of about four hundred target mRNAs (affecting expression of several hundred genes). Friedman et al. estimate that >45,000 miRNA target sites within human mRNA 3′UTRs are conserved above background levels, and >60% of human protein-coding genes have been under selective pressure to maintain pairing to miRNAs. Direct experiments show that a single miRNA can reduce the stability of hundreds of unique mRNAs.
Protein phosphorylation is highly conserved in pathways central to cell survival, such as cell cycle progression relying on cyclin-dependent kinases (CDKs), but individual phosphorylation sites are often flexible. Targets of CDK phosphorylation often have phosphosites in disordered segments, which are found in non-identical locations even in close species. Conversely, targets of CDK phosphorylation in structurally defined regions are more highly conserved. While CDK activity is critical for cell growth and survival in all eukaryotes, only very few phosphosites show strong conservation of their precise positions. Positioning is likely to be highly important for phosphates that allosterically regulate protein structure, but much more flexible for phosphates that interact with phosphopeptide-binding domains to recruit regulatory proteins.

=== Sunlight === There is evidence that β-endorphin is released in response to ultraviolet radiation, either through sun exposure or artificial tanning. This is thought to contribute to addiction behavior among excessive sunbathers and users of artificial tanning despite health risks.
Valuable, temperature-sensitive, or hazardous cargo often require the utmost in system reliability. This type of reliability can only be achieved through the installation of a redundant refrigeration system. A redundant refrigeration system consists of integrated primary and back-up refrigeration units. If the primary unit malfunctions, the secondary unit automatically starts. To provide reliable power to the refrigeration units, these containers are often fitted with one or more diesel generator sets. Containers fitted with these systems may be required for transporting certain dangerous goods in order to comply with the International Maritime Organization’s regulations.
=== Cochlear implants === NASA engineer Adam Kissiah started working in the mid-1970s on what became the cochlear implant, a surgically implanted device that allows people suffering from certain kinds of hearing loss, and who receive little or no benefit from traditional hearing aids. Inspired by his own hearing problems, Kissiah, an electronics instrumentation engineer at NASA, spent 3 years of lunches and evenings in Kennedy Space Center's technical library, studying how engineering principles could affect the inner ear. In 1977, NASA assisted Kissiah in obtaining a patent for the cochlear implant.
=== Opposition to the Peace Preservation Act === In 1878, the Cape Parliament had passed the Peace Preservation Act, which allowed for the confiscation of the firearms of the African population in exchange for a monetary compensation. Sprigg decided that its implementation should extend to the Basuto, after witnessing 7,000 Basuto cavalrymen perform maneuvers during the course of Moorosi's uprising. This was announced during a pitso (formal assembly) attended by some 6,000 to 10,000 Basuto. Soon afterwards he also declared that the Quthing region would be confiscated by the Cape for white settlement. At the time almost half of all Basuto men owned a firearm. Many had worked in railway construction and the diamond mines in Griqualand West with the express purpose of purchasing modern breech-loading and smoothbore rifles. As a result, the Basuto became the best-armed tribe in southern Africa. For the Basuto gun confiscation was unacceptable, not only due to their high value, but also due to the necessity to defend their land and cattle in an environment where there was no guarantee of protection from the colonial authorities. For the Basuto, guns were a symbol of manhood, and to be disarmed was seen as being reduced to the status of a child. In April 1879, the Cape Colony doubled the hut tax in Basutoland to one £ per hut. In June 1880, Letsie I dispatched a three-man delegation to the Cape Parliament as it was deliberating the annexation of Quthing and the question of Basuto disarmament.

Gus Sackey (David Jonsson) is a new grad at Pierpoint, and one of the series' protagonists during the first two series. He is initially assigned to the Investment Banking Division (IBD) and transferred to the CPS desk after the death of his colleague Hari Dhar. Gus is openly gay and hails from an elite background, having graduated literae humaniores from both Eton College and the University of Oxford. Despite his upper-class upbringing, Gus is shown to be humble and morally principled, and feels undervalued within Pierpoint's cutthroat work culture. In series 1, Gus struggles to find a clear role after his team is dissolved and repeatedly alienates senior staff through his bluntness and frustration, including London office head Sara Dhadwal and his manager Clement Cowan. He also rekindles an affair with his former classmate Theo Tuck. On RIF day, Gus delivers a deliberately unserious presentation—beginning by reading from a note written as part of a bet—before walking out, and is not offered a permanent position. In series 2, Gus re-emerges working in politics after striking up a connection with MP Aurore Adekunle, who hires him as an aide. Gus feels fulfilled by the work—which largely involves speaking with everyday constituents about their struggles—despite his sister Sadie's objections. While working for Aurore, Gus begins a relationship with Leo Bloom, the son of hedge fund manager Jesse Bloom, and later leaks confidential government information about the approval of Amazon’s acquisition of pharmaceutical company FastAide to Harper.
=== Teat flow rate === Teat characteristics can also have important implications for infant's sucking pattern and milk intake. Milk flow rate is defined as "the rate at which milk moves from the bottle nipple into the infant's mouth during bottle-feeding." Characteristics such as the shape of the nipple and the way it is perforated may impact flow rate and the coordination of sucking, swallowing and breathing during feeding. Unfortunately, categorization and labeling of teats to indicate flow rate is neither standardized nor consistent. There is significant variability between and within brands and models. In one study, nipples labeled "Slow" or "Newborn" (0–3 months) had flow rates ranging from 1.68 mL/min to 15.12 mL/min."The name assigned to the nipple type does not provide clear information to parents attempting to choose a nipple". This may be of extra concern in the case of fragile infants. Specialized teats are available for infants with cleft palate.
== Early career == Starting in the 1970s Smith published peer-reviewed papers on several subjects, including mass spectrometry, ion cyclotron resonance mass spectrometry, ion-molecule reactions, molecular assemblies, and supercritical fluid solutions. This early work has led him to be considered an internationally recognized expert in mass spectrometry and separation techniques, and his research has led to advancements in instrumentation for the medical and environmental analysis fields, as well as biological research. In the medical field, Smith's work has produced benefits in the areas of drug testing, analysis of pharmaceuticals and medical diagnostics in the clinical arena. His most successful invention has been the combination of capillary electro-phoresis with mass spectrometry. By the end of the 1990s, Smith's achievements included the electrodynamic ion funnel and a micro-dialysis device for the rapid purification of samples analyzed using mass spectrometry. Other notable contributions have been in the fields of supercritical fluids and related reverse micelle phenomena. On August 22, 2000, Smith demonstrated and patented the electrodynamic ion funnel for highly efficient capture and focusing of ions in gases. He applied it for increasing the sensitivity of ESI-MS. His group has continued to refine and extend ion funnel technology, which is now widely applied in mass spectrometry and ion mobility instrumentation.

It is a synthetic peptide modeled on growth hormone-releasing hormone. The molecule is used in research on pituitary growth hormone secretion. It differs from the natural hormone through several stabilizing substitutions.
The DAC form carries a maleimide group that binds albumin and extends circulation time. The non-DAC form lacks this group and clears much faster. Both share the same receptor-binding core.
It targets the growth hormone-releasing hormone receptor on pituitary somatotroph cells. Activation raises cyclic AMP and promotes growth hormone release. Normal feedback pathways remain part of the response.
The version carrying the affinity complex bears a maleimide group that binds serum albumin, which extends its circulation time to several days. The version without it lacks this group and clears within roughly half an hour. The two are chemically related but behave very differently once in the body.
